MBIO 3032 Bacteriology Lab Exam I study guide

 

You should be able to:

·        List and follow the laboratory safety rules and procedures

·        Define all terms in bold face type in lab manual

·        Answer all questions and critical thinking questions in lab reports

·        Identify the parts of a microscope and their functions

·        Differentiate between brightfield, darkfield, and phase contrast microscopy (how they work and what their primary use is)

·        Distinguish between true motility and Brownian motion

·        Explain the purpose for and basic mechanisms of staining (smear prep, fixation)

·        Explain the mechanism and purpose of the following staining techniques:

a.     Negative stain

b.    Gram stain (including purpose and outcome of each step)

c.     Acid-fast stain

d.    Endospore stain

e.     Flagella stain

·        Explain the function of an endospore and the conditions under which they are formed

·        List, describe, and identify the 4 types of flagella arrangements

·        Explain the rationale for aseptic technique and give examples of practices in the lab

·        Differentiate among the following culture media: broth, agar slant, agar deep

·        Describe streak plate, spread plate, and pour plate and their purposes

·        PERFORM and identify the following:

a.     Simple stain

b.    Negative stain

c.     Gram stain

d.    Endospore stain

e.     Streak plate

·        Explain serial dilution, its use and calculation of colony-forming units (CFU)

·        Differentiate between selective and differential media and give examples (MSA, EMB, etc.)  - be able to explain why each example given is selective and/or differential

·        Differentiate between oxidative and fermentative catabolism

·        Explain the purpose and results expected for these biochemical tests:

  1. Starch hydrolysis

  2. Carbohydrate Fermentation

  3. MR-VP test

  4. Urease test

  5.  MIO medium (3 uses)

  6. Indole test

  7. Phenylalanine deaminase test

  8. Nitrate reduction

·        Describe the ways amino acids are catabolized (decarboxylation, deamination)

·        Explain the derivation of H2S in amino acid decomposition and the test to detect it

·        Explain the conditions for growing: obligate aerobes, obligate anaerobes, facultative anaerobes, aerotolerants, and microaerophiles

·        Describe the 3 ways to culture anaerobes

·        Identify the 4 phases of growth and interpret growth data (including determination of generation time, identifying and defining minimum, optimum, maximum growth temperature)

·        List and describe the 5 categories of bacteria as classified by growth temperature

·        Explain the effect of temperature on bacterial growth
